2009-2013年,从青海湖裸鲤内脏器官中共分离获得6株发光细菌,编号依次为Q1、Q2、Q3、Q4、Q5、Q6.这6株发光细菌细胞壁革兰氏染色均为阴性,细胞为杆状,大小为(0.5~0.8)μm×(1.0~1.5)μm,具有单极生鞭毛,有运动性.氧化酶和接触酶均呈阳性反应,能发酵葡萄糖产酸;吲哚试验和V-P试验均为阳性反应,能分泌淀粉酶、明胶酶、DNA酶,能将硝酸盐还原为亚硝酸盐.最适生长温度为20~25℃,最适生长pH为8.5~9.0.16S rDNA基因序列分析结果表明,6株发光菌与其亲缘关系最近的属为Vibrio(弧菌属),该属内和此次实验的6株发光菌亲缘关系最近的种Vibrio anguillarum(鳗弧菌)的16S rDNA序列相似值为99.57%.Biolog GenⅢ鉴定结果显示,该实验的6株发光菌在有氧条件下,在提供的71种碳源中,只能利用其中的30种,其中,糖类12种,氨基酸类5种,羧酸类4种,聚合物类2种,其他类7种.结合6株发光菌的形态学特征、生理生化特性、Biolog Gen III鉴定结果和分子生物学特性,将菌株鉴定为Vibrio anguillarum.该菌所具有的发光特性为首次报道.  相似文献

107 strains producing protease were screened from 260 strains of Antarctic psychrophilic bacteria, among which proteolytic activity of five strains was more than 45 U ml^-1. The 16S rRNA gcne sequences homology and phylogcnetic analysis of five Antarctic psychrophillc bacteria showed that NJ276, NJS-9, NJ16-70,NJ345 belonged tO the described genus Pseudoalteromonas and NJ341 belonged to the genus Colwellia. The growth and the protease characteristic of four Antarctic psychrophilic bacteria had been studied, and the result showed that the 6ptimal temperature for growth and protease-produeing of four strains was about 10℃. Their growth and protease-produeing were still high during incubatlng 2-5 days. The maximum proteolytic activity occurred at pH 9 for four Antarctic psychrophilic bacteria. The optimal temperature of protease action of both strains NJ276 and NJ5-9 was about 50℃, however, the optimal temperature of protease aetlon of both strains NJ341 and NJ345 was about 40 ℃, and their proteolytic activity under 0℃ exhibited nearly 30% of the maximum activity, but their thermal stabilities were weaker. These results indicated that proteases from NJ341 and NJ345 were low-temperature proteases.  相似文献

Both laboratory experiments and numerical modelling were conducted to study the biodegradation and transport of benzene–toluene–xylenes (BTX) in a simulated semi‐confined aquifer. The factors incorporated into the numerical model include advection, hydrodynamic dispersion, adsorption, and biodegradation. The various physico‐chemical parameters required by the numerical model were measured experimentally. In the experimental portion of the study, BTX compounds were introduced into the aquifer sand. After the contaminants had been transported through the system, BTX concentrations were measured at 12 equally spaced wells. Subsequently, microorganisms obtained from the activated sludge of a sewage treatment plant and cultured in BTX mixtures were introduced into the aquifer through the 12 sampling wells. The distribution data for BTX adsorption by the aquifer sand form a nonlinear isotherm. The degree of adsorption by the sand varies, depending on the composition of the solute. The degradation time, measured from the time since the bacteria were added to the aquifer until a specific contaminant was no longer detectable, was 35–42 h for BTX. The dissolved oxygen, after degradation by BTX compounds and bacteria, was consumed by about 40–60% in the entire simulated aquifer; thus the aerobic conditions were maintained. This study provides insights for the biodegradation and transport of BTX in aquifers by numerical modelling and laboratory experiments. Experimental and numerical comparisons indicate that the results by Monod degradation kinetics are more accurate than those by the first‐order degradation kinetics. 从不同养殖场患病暗纹东方鱼屯(Takifaguobscurus)的鳃、肠、肝、肌肉及溃疡部位分离到9株菌株,经人工感染致病性试验,其T-22、T-11、J-2、H-3、H-5表现出较强的致病性,出现症状与自然发病鱼相同。通过对菌体形态特征、培养特性和API20E细菌生化快速鉴定试条的测定,分别鉴定为温和气单胞菌(Aeromonassobria)、豚鼠气单胞菌(Aeromonascaviae)、嗜水气单胞菌(Aeromonashydrophila)、类志贺邻单胞菌(Plesiomonasshigelloides)、霍乱弧菌(Vibriocholerae)。证实这5株菌为致病菌。药敏试验结果表明9种药物对温和气单胞菌、豚鼠气单胞菌、霍乱弧菌均有较好的抑菌效果;嗜水气单胞菌表现出明显的耐药性。氟嗪酸、环丙沙星、复合磺胺对类志贺邻单胞菌有一定抑菌效果。  相似文献

为了解九孔鲍养殖环境中细菌生理菌群的生态作用,对分离自养殖水体的33株菌株的产酶能力和种类进行了研究。结果表明,具有分泌胞外蛋白酶、脂肪酶和纤维素酶能力的菌株分别为39.4%、57.6%和30.3%;在这33株菌中,产至少一种胞外酶的菌株比例高达63.6%。API鉴定结果表明,在有能力分泌胞外蛋白酶、脂肪酶和纤维素酶的菌株中,弧菌菌群所占的比例分别为46%、42%和40%,而其中河流弧菌又占各弧菌菌群比例的83.3%、62.5%和100%。充分阐明弧菌,尤其是河流弧菌为优势菌,在鲍鱼养殖微生物态中起的重要作用。  相似文献

Distribution, variation and impact factors of biomass of bacterioplankton from April to May 1999 in Bohai Sea were studied in DAPI method with epifluorescence microscopy. The biomass in surface waters showed a small day-night variation, varying from 0.13 to 2.51μg/dm^3 with an average of 0.84μg/dm^3. The biomass in bottom waters showed, however, a large variation, changing from 0.15 to 4.18μg/dm^3 with an average of 1.36μg/dm^3. The peak values occurred at 5 and 11 a.m. The bottom water biomass showed a significant correlation with particulate organic carbon (r=0.639, P〈0.05). Heterotrophic bacterioplankton biomass was high in nearshore waters and low in offshore areas with a high biomass zone around Huanghe (Yellow) River mouth, showing the same distribution of nutrients. In vertical distribution, heterotrophic bacteria biomass in bottom waters was higher than that in surface water.  相似文献

A survey was made of the number of the heterotrophic bacteria in surface seawater of Great Wall Bay and its adjacent waters, Antarctica during the period of austral summers from Dec.1993 to Feb. 1994. The result shows that the number of the heterotrophic bacteria in the surface waterof the surveyed area is about 798 CFU. cm-3, ranging from 53 to 4 250 CFU. cm-3. Comparison of thenumbers for three months shows a declining tendency month by month. Comparing with the previous results, this one shows that the heterotrophic bacteria number changes somewhat, but slightly. It is lower than that in nearshore waters but higher than that in the open area. This shows that there is a positive correlative relationship between CFU and NH4- N, which means that heterotrophic bacteria should be an important agent in NH4-N reproduction in the environment surveyed. There is an significant negative correlation both between CFU and NOZ-N, and between CFU and PO4-P. This means that the NOZ-N, PO.-P, which were decomposed and transformed by bacteria, were used rapidly by phytoplankton.  相似文献

以明亮发光杆菌为指示生物,对渔业水域中污染物的急性毒性进行了检测,同时研究了PH值、温度对试验结果的影响。研究表明,发光细菌毒性试验是一种快速、简便的毒性测试方法,在温度20~30℃,pH6.0~9.0条件下所进行的测试,其结果与鱼类毒性试验可以互相替代。  相似文献

Enclosure is commonly used in the restoration of degraded grasslands. However, the effects of enclosure on grassland plant and soil restoration remain controversial, particularly in deserts. To assess the effects of enclosure on desert plants and soil properties, using high throughput sequencing, the differences between plants and soil were systematically analyzed before and after enclosure construction. The soil organic carbon, total nitrogen and total phosphorus contents of the three desert flora increased and decreased, but the difference was not significant; enclosure increased plant height, coverage, aboveground biomass, and species richness by 58.99%, 59.35%, 33.29%, and 51.21%, respectively, in a Seriphidium transiliense formation; by 15.49%, 33.52%, 20.85%, and 5.13%, respectively, in a Haloxylon persicum formation; and by 83.80%, 31.51%, 76.66% and 33.33%, respectively, in an Anabasis salsa formation. For soil bacteria, enclosure significantly increased the average number of operational taxonomic units and Shannon-Wiener index by 12.74% and 2.92%, respectively, under S. transiliense formation and by 17.08% and 3.17%, respectively, under H. persicum formation. However, enclosure had no significant effect on the average number of operational taxonomic units or Shannon-Wiener index under A. salsa formation. Enclosure significantly increased desert plants, soil bacterial diversity, and desert plant community productivity; however, the increase in soil nutrient content was not significant. These results demonstrate that enclosure is effective for restoring desert ecosystems but may have little effect on the soil nutrient content.  相似文献
